LNP Screening Array vs. Micronit Glass Micromixer Chips

By Joshua Gomes

Micronit’s glass micromixer chips are general-purpose components that labs assemble with a holder, tubing, and pumps, while the LNP Screening Array arrives ready to screen LNPs on the liquid handler already in your lab.

LNP Screening Array vs. Micronit Glass Micromixer Chips

Micronit’s Teardrop, Swirl and Double F glass micromixers are general-purpose chips used across chemistry and nanoparticle applications. Using one for LNP formulation means building a fluidic setup around it with a chip holder, tubing and syringe pumps.

The LNP Screening Array is built for LNP formulation screening and runs on the liquid handler already in the lab. Each LNP Screening Array holds eight micromixers in a standard SLAS deck position, and the liquid handler drives every formulation through a sealed pipette interface.

Review published LNP results on standard liquid handlers

Micronit’s product pages publish no LNP protocol or characterization data for these chips. Labs formulatingLNPs on them first have to develop their own mixing parameters and characterization methods before they can screen a lipid library.

The LNP Screening Array has published characterization data for blank LNPs made with SM-102, the ionizable lipid in an approved mRNA vaccine, across five liquid handler platforms. On those platforms, blank SM-102 LNPs measured below 60 nm Z-average diameter and below 0.15 PDI by DLS on a Malvern Zetasizer, with Z-average variability below 3% CV.

Evidence LNP Screening Array Micronit glass micromixer chips
Documented formulation Blank LNPs made with SM-102 No LNP formulation published
Characterization data Z-average diameter, PDI, and Z-average CV by DLS None published for LNPs
Liquid handler platforms tested Five None published

Connect each mixer through a standard pipette tip

Each Micronit chip runs in the Fluidic Connect Pro holder. Micronit specifies its own proprietary ferrules and tubing for the holder and warns that other fittings risk leaks or chip damage. Flow comes from separate syringe pumps, which Micronit sells in a starter kit with the holder and chips.

The LNP Screening Array sits in a downholder in a standard deck position and connects through standard pipette tips on a liquid handler, so there is no tubing, ferrule or pump setup required.

Setup LNP Screening Array Micronit glass micromixer chips
Mixer mount Deck adapter in a standard SLAS deck position Fluidic Connect Pro chip holder
Fluid connection Pipette tip sealed to the pipette interface Proprietary ferrules and tubing
Flow source Liquid handler Separate syringe pumps
Flow rate ratio Fixed at 3:1 by device geometry Set on the pumps for each run
Total flow rate Set in the automation protocol Set on the pumps for each run

Run each formulation through its own micromixer

A Micronit glass chip is reused across a screen, so every changeover needs a cleaning step. Micronit’s cleaning guidance for its glass chips includes a 1 M sodium hydroxide flush, an ultrasonic bath and heating to about 400°C.

On the LNP Screening Array, each formulation runs through its own micromixer, and the liquid handler starts the next formulation as soon as the previous one is collected.

Between formulations LNP Screening Array Micronit glass micromixer chips
Mixer preparation None Flush and clean before reuse
Cleaning materials None Sodium hydroxide, ultrasonic bath, and heat
Operator involvement None between formulations Required at every changeover

Run your first screen during the on-site setup

Micronit offers purpose-built LNP mixers only as custom design and prototyping projects. A lab using the general-purpose chips assembles the holder, fittings, tubing and pumps and develops its own LNP process around them.

The LNP Screening Array comes with an automation protocol configured for your liquid handler, and our applications team installs it and runs the first screen with you on site. LNP Screening Arrays have a three-day lead time.
